All members of the Major cadherins family contain at least five consecutive conserved extracellular cadherin sequence domains, a single transmembrane domain and a cytoplasmic domain that binds to armadillo (ARM) scaffold proteins such as β-catenin and α-importin [1-2].

Subfamilies:
Type I classical cadherins (CDH1-4, -14). The most studied are CDH1 (epithelial/E-cadherin), CDH2 (neural/N-cadherin) and CDH3 (placental/P-cadherin)
Type II classical cadherins (CDH5-12, -18-20, -22, and -24)
7D cadherins (CDH16 and CDH17)
Desmosomal cadherins (DSC1-3 and DSG1-4)
CELSR cadherins (CELSR1-3 which encode Adhesion class GPCRs).

Comment

Cadherin-6 (CDH6) is a type II cadherin that binds intracellularly to catenins via two conserved motifs in its cytoplasmic domain. Cadherin-6 is the target of the investigational anti-tumour antibody-drug conjugate raludotatug deruxtecan [3].
