Used to treat low blood sugar levels in hypoglycemic patients. As glucagon relaxes the smooth muscles of the gastrointestinal tract it is sometimes used in gastric imaging procedures.

Glucagon acts in the liver resulting in the conversion of glycogen stores to glucose. The first step of the process is glucagon-induced activation of the glucagon receptor, triggering a signalling cascade which culminates in the activation of phosphorylase A, the enzyme responsible for the release of glucose-1-phosphate from glycogen polymers.
