Comment Serum from patients with vaccine-induced immune thrombocytopenia and thrombosis (VITT; a serious but very rare adverse response to a chimpanzee adenovirus-based SARS-CoV-2 vaccine [2,8]) activates platelets via the FCGR2A (FcγRIIA) [9]. VITT can be managed with intravenous Ig infusion or plasma exchange, but these are expensive and limited resources. In vitro, pharmacological blockade of FcγRIIA-mediated platelet activation can be achieved by drugs that inhibit the function of COX enzymes, P2Y12, or the Src, Syk and BTK kinases. This indicates that small molecules may have clinical potential as VITT therapies.
