SMARCA2 and SMARCA4 are the central ATPase components of mammalian SWItch/Sucrose Non-Fermentable (SWI/SNF) complexes which play a key role in cell differentiation and proliferation. The human analogue of SWI/SNF is the Polybromo, Brg1-Associated Factors (PBAF) complex which contains the polybromo-1 (PB1) protein along with SMARCA2 and SMARCA4. Genetic lesions of SWI/SNF complexes have been associated with cancer development. Both SMARCA2 and SMARCA4, and PB1 contain a C-terminal bromodomain. Inhibitors targeting these bromodomains are being sought as useful chemical probes for interrogating SWI/SNF mediated processes, potentially offering clues for the development of novel therapeutics.
