Comment: Compound 26 is a macrocyclic inhibitor of apoptosis signal-regulating kinase 1 (ASK1, MAP3K5) [1]. Amongst the analogues designed and tested it is noted for its high level of CNS penetration, making it an ideal lead for examining the effect of ASK1 inhibition on inflammatory demyelination and neurodegeneration. Two previously reported ASK1 inhibitors have shown efficacy in the transgenic mouse SOD1 motor neurone disease model [2].

Immunopharmacology Comments
Inhibition of ASK1 reduces the inflammatory reponse to cellular stress. Peripherally acting ASK1 inhibitors are being investigated for anti-fibrotic activity (e.g. Selonsertib), and centrally acting (CNS penetrant) inhibitors such as compound 26 are predicted to offer potential for the treatment of inflammatory neurodegenerative conditions.
