Comment The VAV1 protein is a hematopoietic-specific signaling protein that plays key regulatory functions in immune cell development and function. It acts as a guanine nucleotide exchange factor (GEF) for Rho/Rac family GTPases and as an adaptor/scaffold protein. Dysregulated VAV1 activity has been linked to hematological malignancies (e.g., T-cell lymphomas) and autoimmune diseases and it is therefore being pursued as a therapeutic target. Targeted degradation of VAV1 protein modulates both T- and B-cell receptor-mediated activity in vitro and in vivo, and disrupts cytokine secretion and immune signaling [2].
