The FSH receptor is the primary target of the approved drug urofollitropin, a recombinamt form of endogenous FSH.

Various cyclic and acyclic α and β aminocarboxamide derivatives (pKd ∼ 8.4 - 5.8) [24], thiazolidine derivatives (pKd ∼ 7.5) [20], biaryl derivatives (pKd ∼ 8.15 - 7.5) [14-15] and thienopyrimidine derivatives (pKd ∼ 7.4 - 6.0) [31] are known to be FSH receptor agonists.

FSH deglycosylated α/β Hs Antagonist 10.0 pKd 13,17
pKd 10.0 [13,17]
Antagonist Comments
Various aminoalkylamide derivatives (pIC50 ∼ 7.0 - 4.3), tetrahydroquinoline derivatives (pIC50 ∼ 6.0) [32] and arylsulfonic acids (pIC50 ∼ 5.5)[6,27,34] are known to be FSH receptor antagonists.
Primary Transduction Mechanisms
Transducer Effector/Response
Gs family Adenylate cyclase stimulation
Comments:  There is some evidence that protein kinase A independent pathways, perhaps through a guanine nucleotide exchange factor, can activate the PI3 kinase pathway.

Comments:  Activation of downstream kinases through protein kinase A dependent pathways have been reported.

Biologically Significant Variants
Type:  Single nucleotide polymorphism.
Species:  Human
Description:  An Ile160 -> Thr mutation resulted in impaired targeting of the receptor to the cell membrane. This mutation has been indentified in patients suffering from partial FSH-resistance syndrome.
References:  8,30
Type:  Single nucleotide polymorphism.
Species:  Human
Description:  A Phe591 -> Ser mutation in the 6th tansmembrane domain results in disruption of cell signalling. This mutant receptor has been found in ovarian sex cord tumors.
References:  18
Type:  Single nucleotide polymorphisms.
Species:  Human
Description:  Two mutations, Thr307 -> Ala and Asn680 -> Ser, are suggested to alter the sensitivity of the receptor to FSH. These mutations have been identified in woman who had given birth to two sets of dizygotic twins.
References:  5,9
General Comments
FSH can bind with high affinity to the extracellular domain of the receptor in the absence of transmembrane domains.
It is not known how the signal is transduced upon FSH binding.
