Comment: GT-1 (LCB10-0200) is a novel antibacterial compound with activity against Gram-negative bacteria [5]. Its struture, in which a siderophore (dihydroxypyridone) is conjugated to a modified aminothiazoylglycyl cephalosporin, was designed to exploit siderophore-dependent pathways for the uptake of iron across the outer membrane of Gram-negative bacteria [5-6].

Bioactivity Comments |
GT-1 demonstrates antibacterial activity against Gram-negative bacteria including Pseudomonas aeruginosa, Klebsiella pneumoniae, Klebsiella oxytoca, Enterobacter aerogenes and Proteus spp. [4-5]. The MIC90 against Pseudomonas aeruginosa is 0.5 μg/ml, with activity (range of 0.03-16 μg/ml) against β-lactamase producing strains also demonstrated [5]. When formulated with GT-055 (LCB18-055), a β-lactamase inhibitor, GT-1 shows enhanced antibacterial activity [2-3]. |
