Summary of Clinical Use
Used in the treatment of depression, including major depressive disorder, bipolar depression, psychotic depression, and involutional melancholia.
Mechanism Of Action and Pharmacodynamic Effects
Maprotiline is a potent norepinephrine reuptake inhibitor. It only weakly inhibits serotonin and dopamine reuptake. Receptor pharmacology: potent antagonist of the histamine H1 and serotonin 5-HT7 receptors; moderate antagonist of the serotonin 5-HT2 and α1-adrenoceptors; weak antagonist of the dopamine D2 and metabotropic acetylcholine receptors.
