Estrone is used to manage peri- and post-menopausal symptoms, importantly estrogen deficiency-induced osteoporosis. Estrone does not appear to have general US FDA or European Medicines Agency marketing approval, although a few national agencies approve this drug.

Estrone binding to estrogen receptor α, creates a transcription factor which binds to estrogen response elements in target gene promoter regions leading to estrogen-regulated protein synthesis.
