Used to treat insomnia, and may be used as sedative. Also used to treat allergic conditions of the upper respiratory tract and skin.

Promethazine blocks the action of endogenous histamine at the histamine H1 receptor. The antiemetic effect may be associated with central anticholinergic actions, with potential activity within the medullary chemoreceptor trigger zone (controls the vomiting center).
