Comment: Ethionamide is a thioamide compound with potent antibacterial activity against mycobacteria. It is a prodrug that requires intracellular activation, by the monooxygenase EthA, in Mycobacterium tuberculosis [1]. Ethionamide is used in the treatment of multidrug-resistant tuberculosis (MDR-TB) and extensively drug-resistant tuberculosis (XDR-TB).

Ethionamide is indicated for the treatment of active tuberculosis when first-line therapies cannot be used because of resistance and/or intolerance. It is is administered orally and in conjunction with other tuberculosis medicines. Ethionamide is on the World Health Organization's Model List of Essential Medicines and was given authorisation by the US FDA in 1965.
