Comment ENL is a chromatin reader that contains a YEATS domain that recognises acylated lysine side chains within the amino-terminal tails of histone H3. It acts as a co-activator within the transcriptional super elongation complex (SEC). ENL has been identified as a driving factor in an aggressive subset of acute leukemia with oncogenic rearrangements of the multiple lineage leukemia (MLL) gene. This suggests that inhibiting ENL's interaction with acylated lysine marks could be leveraged as a therapeutic intervention in these cancers.
