Comment: Nb23-bi is a bivalent nanobody that targets an allosteric domain within the N terminal region of adhesion GPCR ADGRG2 [1]. Nanobody binding induces conformational changes in orthosteric DHEA binding site that promote DHEA induced receptor activation. Nb23-bi reverses the dysfunctions (e.g. orchitis and infertility) caused by gain-of-function ADGRG2 mutants in vivo.
