Comment: Raludotatug deruxtecan (R-DXd; DS-6000a) is an anti-CDH6 monoclonal conjugated to the chemotherapy agent deruxtecan (DXd; a derivative of the topoisomerase I inhibitor exatecan) [1-2]. The antitumour effect of raludotatug deruxtecan is due to intracellular release of the toxic payload; the naked antibody has no anti-tumour activity.

| Raludotatug deruxtecan (R-DXd; DS-6000a) was progressed as a clinical candidate for the treatment of advanced solid tumours. In September 2025 the FDA issued Breakthrough Therapy Designation for raludotatug deruxtecan to treat CDH6-positive gynaecologic cancers (post-bevacizumab treatment) [3]. |
