Comment: Fianlimab (R3767; REGN3767) is a monoclonal antibody that targets the inhibitory immune checkpoint co-receptor lymphocyte activation gene-3 protein (LAG3; CD223) on tumour infiltrating lymphocytes [1]. It was proposed for immunotherapeutic antineoplastic activity.

| Fianlimab (REGN3767) cross-reacts with monkey LAG3 (EC50 0.8 nM for cynomolgus LAG3 expressed on HEK293 cells) [1]. The binding potency is comparable whether LAG3 is recombinantly overexpressed on HEK293 cells, or endogenously expressed on activated primary CD8+ T cells. Fianlimab does not cross-react with rat or mouse LAG3. |
