cedazuridine Small molecule or natural product Approved drug Hs Inhibition 6.4 pIC50 1
pIC50 6.4 (IC50 4x10-7 M) [1]
Description: Inhibition of recombinant hCDA, assessed as decrease in cytidine to uridine formation.
Biologically Significant Variant Comments
Mutations in the CDA gene are associated with decreased sensitivity to the cytosine nucleoside analogue chemotherapy drug cytarabine (cytosine arabinoside, Ara C).
General Comments
Cytidine deaminase (CDA) is an enzyme of the pyrimidine salvage pathway (a subdivision of nucleotide salvage). Salvage returns bases and nucleosides that are formed during degradation of RNA and DNA to the biosynthesis and interconversion pathways. CDA is a homotetrameric enzyme that catalyses the irreversible hydrolytic deamination of cytidine->uridine and deoxycytidine->deoxyuridine. This enzyme is the molecular target of the synthetic nucleoside analogue pharmacoenhancer cedazuridine.
