Comment: GSK3685032 is a first-in-class, non-nucleoside, reversible inhibitor of the epigenetic target DNA-methyltransferase 1 (DNMT1), that was identified from >1.8 million small molecules using a high-throughput enzymatic activity assay [1]. It was developed for potential to treat haematological cancers. GSK3685032 is selective for DNMT1 and does not affect the activities of DNMT3A or DNMT3B. This means that it is less toxic to normal blood cells than less selective hypomethylating agents such as decitabine. GSK3685032 is active in vitro and in vivo, promoting loss of DNA methylation, and inhibiting transcriptional activation and cancer cell growth.
