Comment: PZL-A is a small molecule that is proposed to stabilise the structure and function of mitochondrial DNA polymerase (mtPOLG) [1], with the intention of combating the lethal pathological effects that are caused by mutant POLG and associated mtDNA depletion. PZL-A interacts at the POLG/POLG2 interface, in a position that is distinct from the POLG catalytic sites.
