Immunopharmacology Comments
AZD4144 binds directly to NLRP3 and stabilises the protein in its inactive conformation. Blocking NLRP3 inflammasome formation and activation reduces cleavage of pro-inflammatory IL-1β and IL-18 from their latent forms, and this mechanism mediates the anti-inflammatory action of this class of compounds.
