Comment: ASTX295 is an orally bioactive, short half-life MDM2 proto-oncogene (MDM2, a.k.a. murine double minute 2) antagonist (blocks its interaction with p53) that briefly induces reactivation of p53-mediated transcription, leading to cell cycle arrest and apoptosis in cancer cells with wild-type p53 [2-3]. The chemical structure of ASTX295 was optimised from previously reported antagonists [1]. ASTX295's short duration of action is proposed to limit the risk of marrow suppression/severe thrombocytopenia that arises from extended p53 pathway activation.
