Comment: IL-17C is an IL-17 homolog, secreted by epithelial cells in response to bacterial challenge and inflammatory stimuli and that acts in an autocrine manner to control the innate epithelial immune responses necessary for regulating intestinal inflammation and barrier function [1-2].

Immunopharmacology Comments
Epithelially derived IL-17C acts in an autocrine manner, signaling via an IL-17RA/IL-17RE receptor complex on the epitheilail cells to induce production of proinflammatory cytokines, chemokines and antimicrobial peptides [1].
In light of the potential clinical benefits arising from pharmacological inhibition of IL-17C activity, Galapagos NV, in collaboration with MorphoSys and Novartis, developed the first-in-class anti-IL-17C monoclonal antibody, MOR106, for atopic dermatitis. However, MOR106 development was terminated in 2019 based on an interim analysis for futility that was performed in the Phase 2 IGUANA trial .NCT03568071
