Comment: DKK1 is a secreted protein that inhibits canonical Wnt/β-catenin signalling via interaction with the LRP5/LRP6 co-receptor. It is involved in normal biological processes such as embryonic development and bone homeostasis [5]. In the cancer setting DKK1 promotes an immunosuppressive tumour microenvironment [3,6], and is associated with cancer cell proliferation, invasion, and tumour growth [1,4]. Neutralising DKK1 function is being investigated as an oncology therapeutic strategy.
