P. falciparum phosphatidylinositol 3-kinase (PfPI3K) is the only PI3K that has been identified in the parasite's genome, with sequence analysis and domain organization suggesting that it may be closest to class III PI3Ks, such as VPS34 [1]. The same study found that PfPI3K is involved in endocytosis from the host and trafficking of haemoglobin in the parasite, with inhibition of PI3K causing accumulation of uncatabolized haemoglobin. The PI3K inhibitors wortmannin and LY 294002 have been shown to be effective against PfPI3K [1] but selectivity at the Plasmodium target is likely to be a key issue. |
