Comment: LTV17 is a derivative of the naturally occurring antibacterial lactivicin. It has been designed to have increased potency against Gram-negative bacteria by modification of the lactivicin core scaffold with a phthalimide-conjugated siderophore [2].

| LTV17 exhibits improved activity compared to aztreonam against Gram-negative pathogenic bacteria, including against P. aeruginosa, Acinetobacter baumannii, Klebsiella pneumoniae, and Escherichia coli [2]. It is also active against Stenotrophomonas maltophilia (MIC ≤, 62.5 ng/ml), a Gram-negative species that forms biofilms and that can be resistant to antibacterial drugs [1]. In addition, LTV17 exhibits resistance to inactivation by β-lactamase enzymes [2]. LTV17 inhibits Pseudomonas aeruginosa PAO1 penicillin-binding protein 1A (PBP1a) with an IC50 of 30 nM and PBP3 with an IC50 of 46 nM [2]. |
