Comment: GSK8612 is a selective inhibitor of Tank-binding kinase 1 (TBK1) [2]. TBK1 is a noncanonical member of the IκB kinase (IKK) family that is involved in regulating signalling pathways in several physiological processes including innate immunity [1,3], oncogenesis, energy homeostasis, autophagy, and neuroinflammation. TBK1 is activated downstream of TLR3/TLR4 ligand engagement, via the adaptor protein TRIF. This subsequently results in IRF3 phosphorylation and culminates in IFNβ secretion from cell types including dendritic cells and macrophages. Dysregulated TBK1 activity is associated with autoimmune disorders, hence pharmacological inhibitors are considered for the treatment of (auto)inflammatory diseases.
