Comment: MIDD0301 is a positive allosteric GABAAR modulator [1]. MIDD0301 targets specific GABA receptor subtypes (α4- and α5-containing receptors) that are selectively expressed in airway smooth muscle, airway epithelium, and inflammatory cells [2-3], and in this way offers the potential to alleviate asthma pathophysiology with a reduced likelihood of causing systemic side effects. It acts both as a bronchodilator and as an anti-inflammatory agent.

Bioactivity Comments
MIDD0301 potentiates transmembrane currents in CD4+ T cells isolated from asthmatic mice in the presence of GABA [1]. MIDD0301 exhibits very limited distribution to the brain so has restricted potential to cause central nervous system adverse effects.
An EC50 value of 17 nM is reported by Forkuo et al. (2018) but this was established in ex vivo mouse CD4+ T cells, so does not specify GABAA receptor specificity [1], and we therefore have no primary molecular target interaction for MIDD0301. Patch clamp experiments indicate that MIDD0301 potentiates the GABA response by GABAA receptors containing α1, α2, α3, α4 and α5 subunits (exogenously expressed with β3γ2 subunits) [1].
