Tricyclic antidepressants inhibit reuptake of serotonin and norepinephrine by their respective transporters, SERT (SLC6A4) and NET (SLC6A2), and in addition anatgonise many neurotransmitter receptors. Inhibition of SERT and NET increases synaptic neurotransmitter concentrations which enhances neurotransmission [1,4]. Antagonism of receptor activation may have clinical efficacy but may also contribute to the drug's side effects. See the DrugBank link for further details.
