Comment: This is an orally active RIPK2 inhibitor phosphate ester prodrug [1-2]. It is rapidly cleaved by alkaline phosphatases on contact with intestinal tissue and in whole blood. The prodrug formulation was designed to limit systemic exposure, and to focus activity at the preferred site of action in gut tissue.

Immunopharmacology Comments
The in vivo efficacy of inhibitor 3 was evaluated in a murine 2,4,6-trinitrobenzenesulfonic acid (TNBS) induced colitis model. Results from this model validate the hypothesis that inhibition of the RIPK2/NOD2 pathway blocks inflammatory cytokine production and reduces the gastro-intestinal inflammation that is associated with intestinal epithelial disruption.
