Fosmidomycin is under clinical development as a treatment for uncomplicated P. falciparum malaria. Fosmidomycin provides effective initial clearance of asexual blood stage parasites but concerns have arisen regarding recrudescence rates when the compound is used as a monotherapy in the treatment of uncomplicated P. falciparum malaria [2]. Potential Target/Mechanism Of Action: Fosmidomycin is believed to block the non-mevalonate (MEP) pathway of isoprenoid precursor biosynthesis by inhibiting the second enzyme in the pathway, 1-deoxy-D-xylulose 5-phosphate reductoisomerase (DXR) [1]. |
