Comment: SD-36 is a STAT3 PROTAC degrader [1-2]. It simultaneously binds to the SH1 domain of STAT3 and recruits the cereblon E3 ubiquitin ligase, to promote proteasomal degradation of STAT3. STAT3 degradation is a novel anti-cancer strategy, as an alternative to pharmacological inhibtion.

Bioactivity Comments
SD-36 inhibits proliferation of leukemia and lymphoma cell lines that contain high pSTAT3 in vitro, and is active in the Molm-16 xenograft tumour model [2]. The DC50 value for STAT3 degradation is 60 nM in Molm-16 cells.
