Bioactivity Comments
VX15/2503 binds soluble and cellular SEMA4D of human, rabbit, cynomolgus macaque, marmoset, rhesus macaque, rat and mouse origin and blocks binding of SEMA4D to all three of its known receptors, PLXNB1, PLXNB2, and CD72 [1]. Epitope mapping suggests that VX15/2503 binds a functionally important region of SEMA4D, and may sterically hinder SEMA4D-plexinB1 interactions and/or inhibit SEMA4D homodimerisation (required for semaphorin-plexin signalling). VX15/2503 does not cause cytokine storm, a liability factor for human therapeutic antibodies [1].
