Comment: Puquitinib is an orally active, PI3 kinase inhibitor that induces autophagy by inhibiting the PI3K/AKT/mTOR signaling pathway. Puquitinib is selective for p110δ relative to other PI3K class I enzymes. It is a clinical lead developed for antitumour activity [3].

In AML xenografts combination of puquitinib with cytotoxic drugs (especially daunorubicin) produced synergistic, more potent antitumour activity compared with each agent alone [3]. A kinase selectivity screen found little or no puquitinib inhibitory activity against a panel of protein kinases at a concentration of 1 μM [3]. |
