Comment: ABBV-154 is a investigational antibody-drug conjugate (ADC) in which a glucocorticoid receptor modulator (GRM) payload is linked to the anti-TNαF antibody adalimumab[1]. It is very similar to ABBV-3373 but has a phosphate prodrug version of the GRM to provide improved stability. ABBV-154 was designed to modulate TNF-mediated inflammatory pathways by delivering the glucocorticoid directly into activated immune cells that express membrane-bound TNFα, as a treatment for immune-mediated diseases such as rheumatoid arthritis.
