Mechanism Of Action and Pharmacodynamic Effects
Emibetuzumab blocks the receptor for hepatocyte growth factor (HGF), which is the receptor tyrosine kinase MET, without causing any agonist-like activation of the receptor. In tumour cells MET can be activated by ligand-dependent or ligand-independent (eg receptor dimerisation in MET over-expressing tumours) mechanisms, both of which result in activation of downstream signalling pathways known to be involved in tumor cell proliferation and resistance [2,4-5]. Emibetuzumab blocks this signalling cascade [3].
