Immunopharmacology Comments
RORγt expression is highly restricted to the thymus, where it acts as a crucial regulator in the differentiation of human Th17 cells [1,4,9]. Th17 cells produce IL-17cytokines (preferentially IL-17A and IL-17F) which are central drivers of pathogenesis in a number of autoimmune diseases (e.g. IBD [11], psoriasis [3,10], RA [7], MS [5], COPD and asthma) [2,8,13]. Deletion of RORγt impairs Th17 differentiation in murine and human cells, and RORγt knockout mice do not develop the expected pathologies in various autoimmune disease models. Hence inhibition of RORγt activity is considered as a valid therapeutic mechanism for the identification of novel immunosuppressive agents.
