Comment: CXCL4 is a C-X-C motif chemokine first identified in megakaryocytes and platelets. It is a potent antiangiogenic chemokine and also promotes inflammatory fibrosis [3]. It can be used as a prognostic marker for the fibrosing autoimmune disease systemic sclerosis (scleroderma) [2]. A CXCR3 splice variant, CXCR3B, is a functional receptor for CXCL4 [1].

CXCL4 appears to be the predominant ligand for CXCR3B, in comparison to the CXCR3 ligands CXCL9, -10 and -11. CXCL4 displaces binding of 125I-CXCL10 to CXCR3B with an IC50 of 7.46 nM, and displaces 125I-CXCL4 binding with an IC50 of 1.85 nM [1]. |
