Comment: CXCL4 is a C-X-C motif chemokine first identified in megakaryocytes and platelets. It is a potent antiangiogenic chemokine and also promotes inflammatory fibrosis [3]. It can be used as a prognostic marker for the fibrosing autoimmune disease systemic sclerosis (scleroderma) [2]. A CXCR3 splice variant, CXCR3B, is a functional receptor for CXCL4 [1].

Plasmacytoid dendritic cell-derived CXCL4 is involved in the pathogenesis of systemic sclerosis. In vitro, CXCL4 induces the secretion of endothelin 1 by human endothelial cells, and in vivo (in mice) promotes expression of highly proinflammatory CCL2, infiltration of inflammatory cells to sites of CXCL4 injection and induces dermal thickening [2]. |
