Synonyms: DNA-PK inhibitor II | LY-293646 | NU 7026 | NU7026
Compound class:
Synthetic organic
Comment: NU-7026 is a potent and selective, ATP-competitive inhibitor of DNA-dependent protein kinase (DNA-PK) [5].
Ligand Activity Visualisation ChartsThese are box plot that provide a unique visualisation, summarising all the activity data for a ligand taken from ChEMBL and GtoPdb across multiple targets and species. Click on a plot to see the median, interquartile range, low and high data points. A value of zero indicates that no data are available. A separate chart is created for each target, and where possible the algorithm tries to merge ChEMBL and GtoPdb targets by matching them on name and UniProt accession, for each available species. However, please note that inconsistency in naming of targets may lead to data for the same target being reported across multiple charts. ✖ |

Mechanism Of Action and Pharmacodynamic Effects |
DNA-dependent protein kinase (DNA-PK) activity is essential for repair of DNA double-strand breaks [7]. Inhibition of DNA-PK sensitises cells to chemo- or radio-therapy agents [4,6], leading to increased DNA damage being induced by these types of cancer therapies. DNA-PK is a member of the phosphatidylinositol (PI) 3-kinase related kinase (PIKK) family involved in the signalling of cellular stress responses, which also includes the mammalian proteins ATM, ATR, mTOR and hSMG1 [4,8]. NU-7026 is 60-fold selective for DNA-PK over other PIKK family members. |
